Analyses of the mitochondrial cytochrome b gene (1140 bp) showed that Dremomys lokriah , D. pernyi , D. pyrrhomerus , D. rufigenis and D. gularis all are separate species. Dremomys pyrrhomerus showed 8.5% sequence variation from D. rufigenis, and the level of estimated sequence divergence observed among D. gularis , D. lokriah and D. pernyi was > 11%. With Tamiops and Callosciurus as the outgroup taxa, in both maximum likelihood and Bayesian analyses, the five Dremomys species formed one strongly supported monophyletic group and D. pyrrhomerus is closely related to D. rufigenis . The derived divergence times and fossil record suggested that the present geographical distributions of Dremomys owe much to the uplifting of the Himalayas and the successive glacial and interglacial in the Pliocene–Pleistocene.  相似文献

Along with the development and integration of molecular genetics and quantitative genetics, many quantitative trait locus (QTL) mapping studies have been conducted using different mapping populations in various crop species. Existing QTLs can be used for marker-assisted breeding and map-based cloning, whereas the false-positive QTLs are no use. The purpose of this study is to evaluate the suitability of different mapping procedures for data from different genetic models. In this study, four types of recombinant inbred lines (RILs) with different genetic models, viz. additive QTLs (Model I), additive and epistatic QTLs (Model II), additive QTLs and QTL × environment interaction (Model III), additive, epistatic QTLs and QTL × environment interaction (Model IV), were simulated by computer. Six types of QTL mapping procedures, viz. CIM, MIMF, MIMR, ICIM, MQM and NWIM, on four kinds of QTL mapping software, viz. WinQTL Cartographer Version 2.5, IciMapping Version 2.0, MapQTL Version 5.0 and QTLnetwork Version 2.0, were used for screening QTLs of the simulated RILs. The results showed that different mapping procedures have different suitability for different genetic models. CIM and MQM can only screen Model I data. MIMR, MIMF and ICIM can only screen Model I and Model II data. NWIM can screen all four models’ data. It can be concluded that different genetic models’ data have different most suitable mapping procedures. In practical experiments where the genetic model of the data is unknown, a multiple model mapping strategy should be used, that is a full model scanning with complex model procedure followed by verification with other procedures corresponding to the scanning results.  相似文献

Deep‐sea lobsters previously assigned to the family Thaumastochelidae Bate, 1888, the thaumastocheliforms, have very distinctive, greatly unequal first chelipeds, with the right side extremely elongate and pectinate, and in having short, quadrate pleonal pleura. Despite interesting morphology and a long taxonomic history, the phylogeny of the group has received little detailed analysis. Here, we conduct a species‐level phylogenetic analysis of the thaumastocheliforms based on morphological and molecular data (three mitochondrial genes: COI, 16S rDNA and 12S rDNA; two nuclear protein‐coding genes: H3 and NaK) to robustly reconstruct their evolutionary history and estimate divergence times. Separate and combined analyses of all data sources support thaumastocheliform monophyly, but as a clade deeply nested within the Nephropidae supporting recent synonymy of Thaumastochelidae with Nephropidae. Combined and molecular‐only analyses support generic monophyly of all three thaumastocheliform genera and Dinochelus as sister to Thaumastochelopsis, fully corroborating the current, morphology‐based taxonomy. In contrast, Thaumastocheles is recovered as paraphyletic in morphology‐only analyses owing to minimal character support. The Cretaceous–Paleogene Oncopareia was recovered as a stem‐lineage thaumastocheliform. The fossil record indicates that the thaumastocheliforms once lived in shallow, continental shelf depths, but moved into deeper water in the Cenozoic where they occur today. The thaumastocheliforms originated in northern Europe during the Mid‐Late Cretaceous and later dispersed westwards to the south‐eastern Pacific through the western Atlantic and eastwards to the western Pacific through the Indian Ocean. Thaumastochelopsis can be considered the most derived thaumastocheliform genus based on the degree of structural reduction relative to other thaumastocheliforms, its remote geographical occurrence (Australia) from the hypothesised place of origin (northern Europe) and its more recent estimated divergence than other genera (28 Mya for the MRCA of extant species of the genus).  相似文献

The dengue fever epidemic in Guangzhou may have been affected by the Coronavirus Disease 2019 (COVID-19) pandemic. The number of dengue cases dropped drastically in 2020, and there have been only 2 local cases, suggesting that dengue has not become endemic in Guangzhou.

Guangzhou is located on the southeast coast of China and is the country’s third largest city. Since 1978, outbreaks of dengue fever have occurred intermittently in this city. In the past decade, the number of reported dengue cases reached more than 1,000 in 2013, 2014, 2018, and 2019, with 37,385 cases reported in 2014 alone. Therefore, dengue fever is a major public health concern in Guangzhou, and there is a continuing argument that it is endemic in Guangzhou [13].The numbers of dengue cases from 2017 to 2020 are shown in Table 1. In 2020, the total and local case numbers dropped dramatically compared to the previous years. With a high proportion of imported cases (n = 32, 94.12%), the proportion of local cases (n = 2, 5.88%) was considerably low in 2020. All the prevention and control strategies for dengue, including issuing public education messages, preventing further mosquito bites in patients, cleaning vector breeding sites, and using pesticides, were similar during these years. Additionally, dengue, as a mosquito-borne viral infectious disease, is closely related to mosquito density. The mosquito ovitrap index (MOI), which is the proportion of positive mosquito ovitraps, is usually used to indicate mosquito density. The MOI in 2017, 2018, and 2019 was 7.073 ± 1.016, 9.657 ± 1.307, and 8.464 ± 0.961, respectively. The average MOI was 8.398 ± 0.648 from 2017 to 2019 in Guangzhou. The MOI in 2020 was 7.135 ± 0.786, which remained at the median risk level. Therefore, the abnormal decline in dengue cases could not be attributed to the change in mosquito density in 2020.Table 1Numbers and percentages of dengue cases from 2017 to 2019.

Open in a separate windowIn 2020, the 14-day quarantine in a designated hotel for international travelers to curb the spread of Coronavirus Disease 2019 (COVID-19) was an important public health intervention. People had to remain indoors except for medical care needs. All imported dengue cases were identified during their quarantine periods. No secondary case related to the imported cases was reported. This may be because Aedes albopictus, which is the major vector of dengue in Guangzhou, bites aggressively during the day outdoors. The chance of being bitten by A. albopictus was reduced by staying all day indoors. Moreover, some research revealed that viremia occurred 6 to 18 hours before symptoms appeared and lasted as long as 12 days [4]. After the 14-day quarantine, viremia had almost subsided. Therefore, imported dengue cases were unlikely to be transmitted. The impact of imported dengue cases was limited by the quarantine, which provided a rare opportunity to identify the local epidemic.The epidemiology investigation showed that the 2 local cases, who were living in the same building, had no travel history outside Guangzhou in 2020 and had symptoms successively. Two dengue virus serotype 2 (DENV-2) strains were isolated from them. The envelope gene sequences were obtained and deposited in GenBank under accession numbers MW295818 and MW345921. Reference sequences, which were downloaded from GenBank, and sequences of Guangzhou strains identified in the previous years, were used to construct a phylogenetic tree. The 2 isolated strains in 2020 were identical. The tree (Fig 1) shows that the 2 strains belonged to the Malaysia/Indian subcontinent genotype, which was the prevailing genotype in Guangzhou [5]. However, they were neither identical with nor derived from the Guangzhou strains obtained from the previous years. Using the Basic Local Alignment Search Tool in GenBank, the 2 strains were found to be highly similar to those identified in Zhejiang (China), Singapore, and Guangdong (China) in 2017. Hepatitis B virus (HBV) pre-S2 mutant can induce hepatocellular carcinoma (HCC) via the induction of endoplasmic reticulum stress to activate mammalian target of rapamycin (MTOR) signaling. The association of metabolic syndrome with HBV-related HCC raises the possibility that pre-S2 mutant-induced MTOR activation may drive the development of metabolic disorders to promote tumorigenesis in chronic HBV infection. To address this issue, glucose metabolism and gene expression profiles were analyzed in transgenic mice livers harboring pre-S2 mutant and in an in vitro culture system. The pre-S2 mutant transgenic HCCs showed glycogen depletion. The pre-S2 mutant initiated an MTOR-dependent glycolytic pathway, involving the eukaryotic translation initiation factor 4E binding protein 1 (EIF4EBP1), Yin Yang 1 (YY1), and myelocytomatosis oncogene (MYC) to activate the solute carrier family 2 (facilitated glucose transporter), member 1 (SLC2A1), contributing to aberrant glucose uptake and lactate production at the advanced stage of pre-S2 mutant transgenic tumorigenesis. Such a glycolysis-associated MTOR signal cascade was validated in human HBV-related HCC tissues and shown to mediate the inhibitory effect of a model of combined resveratrol and silymarin product on tumor growth. Our results provide the mechanism of pre-S2 mutant-induced MTOR activation in the metabolic switch in HBV tumorigenesis. Chemoprevention can be designed along this line to prevent HCC development in high-risk HBV carriers.  相似文献

Summary Immunohistochemical and ultrastructural techniques have been used to demonstrate glial fibrillary acidic protein (GFAP) immuno-positive cells in the adult toad spinal cord. Two types of GFAP-immunoreactive cells were observed: ependymocytes and radial astrocytes. GFAP-positive ependymocytes were scarce and contained the immunoreactive product in their processes. They showed intermediate filaments in the basal pole and in their processes when studied with the electron microscope. These immuno-positive ependymocytes represent the tanycytic form of ependymal cells because their processes ended at the subpial zone. The radial astrocytes showed a more intensive immunoreactive product in somata and processes when they were located far away from the ependymal layer. Cell bodies and processes were also associated with blood vessels, but most of the processes ended at the subpial zone forming a continuous subpial glia limitans. The GFAP-positive processes, which form this subpial glia limitans in the toad spinal cord, belong to both tanycytic ependymocytes and radial astrocytes, whose somata are located in the grey matter. These findings lead us to suggest that both types of GFAP-immunopositive cells might be the functional equivalents of mammalian astrocytes.  相似文献

Adherent granulocytes secrete a granulocyte factor (GF). This factor stimulates hemopoiesis in irradiated mice. The target cell for this hemopoiesis-regulating GF action is probably T lymphocyte. The results indicate that the target cell is hydrocortisone- and cyclophosphamide-sensitive and prove that granulocytes stimulate hemopoiesis in irradiated mice via secreted GF.  相似文献

Pancreatic islets of Langerhans were perifused with Krebs-bicarbonate solution containing glucose (5 and 10 mM). The perifusate was spiked with tetradeuterated hepoxilin A3 and was extracted and analysed by gas chromatography-mass spectrometry using NICI detection. Evidence is presented showing the presence of hepoxilin A3 as the hydrolysis product trioxilin A3. These results demonstrate for the first time that this pathway is active in intact cells; this finding, taken together with our previous evidence that hepoxilins possess insulin secretagogue properties further supports our hypothesis that these products could play a role as endogenous mediators of insulin release.  相似文献
